Mechanism of Action
MT-2 is a non-selective agonist at melanocortin receptors MC1R through MC5R. Binding to MC1R on melanocytes activates Gαs/adenylyl cyclase/cAMP signaling, which phosphorylates CREB and upregulates transcription of the MITF gene — the master regulator of melanogenesis. MITF drives expression of tyrosinase and tyrosinase-related proteins (TRP-1, TRP-2) that catalyze the conversion of L-tyrosine to melanin intermediates (DOPA, dopaquinone) and ultimately eumelanin. MC3R and MC4R activation in hypothalamic nuclei modulates downstream signaling through the leptin-melanocortin axis.
