Mechanism of Action
TB-500 sequesters G-actin monomers via its LKKTETQ binding motif, modulating the G-actin/F-actin equilibrium and influencing cytoskeletal reorganization, cell migration, and organelle trafficking. Beyond actin regulation, Tβ4 interacts with the PINCH-1/ILK/parvin complex involved in integrin-linked kinase signaling and has been shown to modulate NF-κB-mediated inflammatory gene transcription. The N-terminal tetrapeptide fragment (Ac-SDKP) generated by enzymatic cleavage via prolyl oligopeptidase is itself biologically active in regulating fibroblast differentiation.
