Product Overview
SLU-PP-332 is a small-molecule agonist of the estrogen-related receptor (ERR) family, with primary activity at ERRα and ERRγ subtypes. Despite the naming convention, ERRs are orphan nuclear receptors with no significant binding affinity for estrogen. They function as constitutive transcription factors regulating genes involved in mitochondrial biogenesis, oxidative phosphorylation, and fatty acid metabolism.

Mechanism of Action
SLU-PP-332 binds to the ligand-binding domain of ERRα and ERRγ, stabilizing the receptor in an active conformation and enhancing coactivator recruitment (particularly PGC-1α). This promotes transcription of target genes involved in mitochondrial biogenesis (TFAM, NRF1), oxidative metabolism (PDK4, CPT1B, MCAD), and energy substrate utilization. ERR activation drives a transcriptional program that shifts cellular metabolism toward oxidative phosphorylation and fatty acid oxidation over glycolysis.
