Mechanism of Action
MOTS-C activates the AMPK pathway by inhibiting the folate-methionine cycle, specifically targeting the enzyme MTHFD2 in de novo purine biosynthesis. This leads to accumulation of the intermediate AICAR, a known endogenous AMPK activator. AMPK activation subsequently modulates downstream targets including ACC (fatty acid synthesis), mTORC1 (protein synthesis), and GLUT4 translocation (glucose uptake). Under metabolic stress, MOTS-C translocates from the cytoplasm to the nucleus where it interacts with transcription factors (ARE-Nrf2 and FOXO1) to regulate adaptive gene expression programs involved in cellular stress response.
