Mechanism of Action
5-Amino-1MQ competitively inhibits NNMT by occupying the nicotinamide substrate-binding pocket. NNMT methylates nicotinamide (a precursor to NAD+) into 1-methylnicotinamide, effectively diverting nicotinamide away from the NAD+ salvage pathway. By inhibiting NNMT, 5-Amino-1MQ increases intracellular nicotinamide availability for conversion to NAD+ via nicotinamide phosphoribosyltransferase (NAMPT). This results in elevated NAD+ levels, which serve as a critical cofactor for sirtuin deacetylases (SIRT1-7) and poly(ADP-ribose) polymerases (PARPs). NNMT inhibition also reduces SAM consumption, shifting the SAM/SAH ratio and altering cellular methylation dynamics. In adipocyte models, NNMT knockdown or pharmacological inhibition increases energy expenditure gene expression, including uncoupling proteins and PGC-1α targets.
